چکیده

Transplantation of human umbilical cord mesenchymal stem cells (UC-MSCs) has been shown to be effective in treating critical limb ischemia (CLI). However, the mechanism of MSCsmediated improvements in CLI patients, especially with respect to the immuneinflammatory aspects of this disease, is still unknown. In this study, we investigated the changes in T-lymphocyte subpopulations and inflammatory mediators (such as IL-6, IL-10 and TNF-α) in PBMCs from CLI patients after UC-MSCs treatment, as well as the correlation between inflammatory mediators and EPCs. TNF-α serum levels increased at 24 h (p=0.017) after treatment and then decreased at 1 month (p=0.031) compared with that before treatment. IL6 serum levels increased at 24 h (p=0.099) after treatment and then decreased at 1 month (p=0.072) compared with that before treatment. The percentages of CD3+ T, CD3+ CD4+ lymphocytes and NK cells were significantly decreased after UC-MSCs treatment (p=0.002, p=0.012 and p=0.029, respectively). TNF-α (r=-0.42, p=0.0004) and IL-6(r=-0.33, P < 0.0001) were shown to be inversely correlated with the number of circulating EPCs. Our studies have shown that UC-MSCs have anti-inflammatory and immunomodulation properties in CLI and promotes healing of non-healing wounds. Growth suppression effect of human mesenchymal stem cells from bone marrow, adipose tissue, and Wharton's jelly of umbilical cord on PBMCs

Objective(s):Immunosuppressive property of mesenchymal stem cells (MSCs) has great attraction in regenerative medicine especially when dealing with tissue damage involving immune reactions. The most attractive tissue sources of human MSCs used in clinical applications are bone marrow (BM), adipose tissue (AT), and Wharton's jelly (WJ) of human umbilical cord. The current study has compared immu...

Neuroinflammation has a significant role in induce of Multiple sclerosis (MS) many approaches have been used to treat MS, but none of these methods have not been able to fully improve. One of the methods can suppress inflammation and regenerate the nervous system is the use of cell therapy. Using cell therapy in pre-clinic phase can be realized, it's mechanism and potency to suppress neuroinfla...

Mesenchymal Stem Cells: Interactions with Immune Cells and Immunosuppressive-Immunomodulatory Properties

Abstract Background and Objectives Recently, mesenchymal stem cells have attracted much attention in regenerative medicine and cell-based therapies. Mesenchymal stem cells are used in regenerative medicine mainly based on their capacity to differentiate into several cell lineages, low immunogenicity, and in particular their anti-inflammatory and immunosuppressive-immunomodulatory properties. ...

The effect of amniotic membrane extract on umbilical cord blood mesenchymal stem cell expansion: is there any need to save the amniotic membrane besides the umbilical cord blood?

Objective(s): Umbilical cord blood is a good source of the mesenchymal stem cells that can be banked, expanded and used in regenerative medicine.  The objective of this study was to test whether amniotic membrane extract, as a rich source of growth factors such as basic-fibroblast growth factor, can promote the proliferation potential of the umbilical cord mesenchymal stem cells. Materials and ...

P 141: Mesenchymal Stem Cells as Treatment in Neuroinflammatory Disease

Mesenchymal stem cells can be obtained from deferent tissues like adipose tissue, umbilical cord, placenta, skin, bone marrow, etc. These cells have regulatory effects on all types of immune cells such as dendritic cell, natural killers and lymphocytes. Mesenchymal stem cells induce inhibitory phenotypes of Antigen Presenting Cells (APCs) following their activity. They also change T cells pheno...
